固定/ 破膜劑

（一）應用于流式細胞儀檢測臨床全血標本細胞內多項免疫學指標
肝素鈉抗凝的全血樣本，室溫存放，在24 小時內使用。
1. 取抗凝血50ul 放入流式管中。
2. 加入檢測細胞表面分子用的熒光素標記抗體，混和均勻，室溫避光孵育20 分鐘 。
3. 加入1ml 溶血素，振蕩均勻，室溫放置20 分鐘，離心（300g 5 分鐘）去除上清液。
4. 用1ml PBS 重懸洗滌細胞，離心（300g 5 分鐘）去除上清液。
5. 加入300ul 固定/ 破膜劑，振蕩均勻，室溫放置20 分鐘。離心（300g 5 分鐘）去除上清液。
6. 加入胞內細胞因子檢測用熒光素標記抗體，混和均勻，室溫避光孵育20 分鐘 。
7. 用1ml PBS 重懸洗滌細胞，離心（300g 5 分鐘）去除上清液。
8. 上機分析。

（二）應用于流式細胞儀檢測外周血PBMC 的細胞內多項免疫學指標
1. 使用淋巴細胞分離液分離外周血樣本，獲得新鮮PBMC（外周血單個核細胞）后，取1X106的細胞放入流式管中。
2. 加入檢測細胞表面分子用的熒光素標記抗體，混和均勻，室溫避光孵育20 分鐘 。
3. 加入300ul 固定/ 破膜劑，振蕩均勻，室溫放置20 分鐘。離心（300g 5 分鐘）去除上清液。
4. 加入檢測細胞內細胞因子用的熒光素標記抗體，混和均勻，室溫避光孵育20 分鐘 。
5. 用1ml PBS 重懸洗滌細胞，離心（300g 5 分鐘）去除上清液。
6. 上機分析。
